How do I get from Tokyo to Yokohama?

I'm coming to Tokyo next week to cover the Auto Show and I thought I would take a day to visit Yokohama.
I'm staying at the Prince Shinagawa Hotel and I understand I can catch a train to Yokohama from that station. Do you know where I would buy the ticket and what time the last train leaves Yokohama back for Tokyo?

First up, Yokohama is well worth a visit! It's a great area and a different feel to Tokyo.
To answer your question, the easiest line to catch direct from Shinagawa station is the Yokosuka Line (check out this map). The Shinagawa station is at the bottom of the central loop line marked on the map and Yokohama is South on the dark blue line. You can also take the Keihin Tohoku Line it's just a little slower with more stops.
You can buy tickets for either of these lines straight from the hole in the wall ticket machines at the station itself. If you have a pre-paid Suica pass or Pasmo card then you can just put credit on your card and swipe yourself through the ticket barrier. If you need to buy a ticket from the machine, check the map that is nearby (usually overhead). Find Yokohama on the map and then follow the line map to see how much it will cost for a single trip to Yokohama.
If you get stuck then just to go the guard at the gate and say "Yokohama onegaishimasu" - it's very simple but said with a smile will usually get you want without getting you into trouble! The guard can get the ticket for you there.
If you have a Japan Rail Pass then both lines are JR so you can use the pass on either.
